The idea was ambitious: growth hormone burns fat, but it brings everything else along with it — it raises blood sugar, messes with insulin, makes cells multiply. If the fat-burning part is just its tail, then you cut off the tail and keep only that. AOD-9604 is that tail: amino acids 176 to 191.
And it worked — in mice. Obese and lean mice, 14 days of continuous dosing via osmotic pumps. Both growth hormone and AOD-9604 significantly reduced weight gain, with increased fat oxidation and increased plasma glycerol — in other words, real lipolysis 1.
And here's the interesting half. Unlike growth hormone, AOD-9604 didn't cause hyperglycemia and didn't reduce insulin secretion. The reason turned up in the same experiment: it doesn't even compete for the growth hormone receptor, and it doesn't cause cell proliferation 1. Thirteen years later the same thing was confirmed from another angle: it has no effect at all on the WADA doping test for growth hormone 2. The body simply doesn't see it as growth hormone.
That's the good news and the bad news at once. Good: it doesn't bring growth hormone's baggage. Bad: it doesn't do any of what growth hormone does either.
And now the timeline. AOD-9604 entered phase II clinical trials for obesity in the early 2000s 3, and was still listed as in development in 2006 4. Two decades on, it isn't an approved obesity drug anywhere. We're not telling you why — we don't know. We're telling you it didn't make it.
